Impaired Repopulating Ability of Uhrf2−/− Hematopoietic Progenitor Cells in Mice

UHRF proteins catalyze the ubiquitination of target and are involved in regulating gene expression. Some studies reported a reduced expression UHRF2 acute leukemia cells, but role hematopoiesis remains unknown. Here, we generated Uhrf2−/− mice to clarify deletion hematopoiesis. Compared Uhrf2+/+ mice, showed no differences complete blood counts, as well bone marrow (BM) findings spleen weights. Proportions cells progenitor fractions BM were comparable between mice. However, competitive repopulation assays with transplants (BMT), proportions decreased relative all lineages. After second BMT, neutrophils few, while 20–30% T B still detected. RNA sequencing downregulation some genes associated stem-cell function hematopoietic stem/progenitor (HSPCs). Interestingly, trimethylated histone H3 lysine 9 was increased HSPCs cleavage under targets tagmentation assay.
